Loeys-Dietz syndrome (LDS) is a genetic disorder characterized by the enlargement of the aorta, the main blood vessel that carries blood from the heart to the rest of the body. It is a rare, life-threatening condition that affects the body's connective tissue.

Overview[edit | edit source]

Loeys-Dietz syndrome is caused by mutations in the TGFBR1, TGFBR2, SMAD3, TGFB2, and TGFB3 genes. These genes provide instructions for making proteins that are involved in the regulation of cell growth and division, as well as the production of the extracellular matrix, a lattice-like network that provides strength and flexibility to the tissues.

Symptoms[edit | edit source]

The symptoms of Loeys-Dietz syndrome can vary greatly from person to person. However, common symptoms include an enlarged aorta, which can lead to aortic dissection or aneurysm. Other symptoms can include skeletal abnormalities, such as pectus excavatum (sunken chest) or pectus carinatum (protruding chest), scoliosis (curvature of the spine), and joint hypermobility (excessive flexibility of the joints). Some individuals may also have craniofacial abnormalities, such as widely spaced eyes, a cleft palate, or a split uvula.

Diagnosis[edit | edit source]

Diagnosis of Loeys-Dietz syndrome is based on clinical evaluation, detailed patient history, and specialized genetic testing. Imaging studies, such as echocardiography and magnetic resonance imaging (MRI), may also be used to detect an enlarged aorta or other cardiovascular abnormalities.

Treatment[edit | edit source]

There is currently no cure for Loeys-Dietz syndrome. Treatment is focused on managing the symptoms and preventing complications. This may include surgery to repair the aorta, medication to control blood pressure, and regular monitoring of the cardiovascular system. Individuals with Loeys-Dietz syndrome are also advised to avoid strenuous physical activity, which can put additional strain on the aorta.
