PTI-3 is a chemical compound that is used as a pharmacological tool in the study of the Sigma-1 receptor. It is one of the most selective Sigma-1 receptor ligands currently available, and has been used to demonstrate that the Sigma-1 receptor mediates the neuroprotective effects of pregnenolone and DHEA.

Chemistry[edit | edit source]

PTI-3 is a benzomorphan derivative, and is structurally related to the opioid antagonist naloxone. However, unlike naloxone, PTI-3 does not have significant affinity for opioid receptors, and instead binds selectively to the Sigma-1 receptor.

Pharmacology[edit | edit source]

PTI-3 acts as an agonist at the Sigma-1 receptor, and has been shown to produce neuroprotective effects in animal models of neurodegenerative disease. These effects are thought to be mediated by the Sigma-1 receptor, as they can be blocked by the selective Sigma-1 receptor antagonist BD-1063.

Research into the potential therapeutic applications of PTI-3 is ongoing. It has been suggested that Sigma-1 receptor agonists like PTI-3 may have potential in the treatment of neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease, as well as in the treatment of depression and anxiety disorders.
