Arachidonate 5-lipoxygenase, often abbreviated as 5-LO or 5-LOX, is an enzyme that in humans is encoded by the ALOX5 gene. This enzyme plays a crucial role in the biosynthesis of leukotrienes, which are lipid-based molecules involved in inflammation and allergic reactions. The activity of 5-LOX leads to the production of leukotrienes from arachidonic acid, a polyunsaturated fatty acid that is a common component of cell membranes.

Arachidonate 5-lipoxygenase catalyzes the first step in the leukotriene synthesis pathway, which is the conversion of arachidonic acid to 5-hydroperoxyeicosatetraenoic acid (5-HPETE), and subsequently to leukotriene A4 (LTA4). LTA4 can then be converted into other leukotrienes such as leukotriene B4 (LTB4), which is a potent chemoattractant for neutrophils, and the cysteinyl leukotrienes (LTC4, LTD4, LTE4), which are involved in bronchoconstriction and increased vascular permeability.

Clinical Significance[edit | edit source]

The leukotrienes produced by the action of 5-LOX are significant mediators in various inflammatory and allergic conditions, including asthma, rheumatoid arthritis, and inflammatory bowel disease (IBD). Consequently, inhibitors of 5-LOX and the leukotriene pathway have therapeutic potential in treating these conditions. For example, zileuton is a direct inhibitor of 5-LOX used in the management of asthma.

Genetics[edit | edit source]

The ALOX5 gene is located on chromosome 10 in humans and contains multiple exons and introns. Variations in this gene have been associated with differences in the enzyme's activity and expression, which can affect an individual's susceptibility to inflammatory diseases.

Regulation[edit | edit source]

The activity of arachidonate 5-lipoxygenase is regulated by various factors, including its cellular localization and interaction with other proteins such as 5-lipoxygenase-activating protein (FLAP). Calcium ions and phosphorylation events also modulate its activity.

Research into the role of 5-LOX and leukotrienes in disease continues to be an active area of study, with the aim of developing new therapeutic agents that target this pathway. Understanding the detailed mechanisms of 5-LOX regulation and action may lead to improved treatments for a wide range of inflammatory conditions.
