CENPK
CENPK (Centromere Protein K) is a protein that in humans is encoded by the CENPK gene. It is a component of the CENP-A-containing nucleosome-associated complex (NAC) and is involved in the assembly of kinetochore proteins, mitotic progression, and chromosome segregation.
Structure[edit | edit source]
CENPK is a part of the CENP-H/I/K complex, which is essential for the proper function of the kinetochore. The protein structure of CENPK allows it to interact with other centromere proteins, facilitating the recruitment of additional components necessary for kinetochore assembly.
Function[edit | edit source]
CENPK plays a critical role in the formation of a functional kinetochore, which is crucial for accurate chromosome segregation during mitosis. It is involved in the recruitment of other centromere proteins and is necessary for the stability of the CENP-A nucleosome at the centromere.
Clinical Significance[edit | edit source]
Mutations or dysregulation of CENPK can lead to improper chromosome segregation, which is a hallmark of many cancer types. Studies have shown that overexpression of CENPK is associated with poor prognosis in certain cancers, making it a potential target for therapeutic intervention.
Research[edit | edit source]
Ongoing research is focused on understanding the precise molecular mechanisms by which CENPK contributes to kinetochore function and its potential role in cancer progression. Researchers are also exploring the possibility of targeting CENPK in cancer therapy.
