CUL7 is a gene that encodes a protein known as Cullin 7, which is part of the cullin family of proteins. These proteins are involved in the ubiquitin-proteasome system, a critical pathway for protein degradation and regulation within the cell.
Structure[edit | edit source]
The CUL7 protein is a member of the cullin family, which are scaffold proteins that assemble multi-subunit ubiquitin ligase complexes. These complexes are responsible for targeting specific proteins for ubiquitination and subsequent degradation by the proteasome. The CUL7 protein contains several domains, including a cullin homology domain and a C-terminal domain that interacts with other components of the ubiquitin ligase complex.

CUL7 plays a crucial role in the regulation of cell growth and development. It is involved in the ubiquitination of specific substrates, marking them for degradation. This process is essential for maintaining cellular homeostasis and regulating various cellular processes, including the cell cycle, signal transduction, and apoptosis.
CUL7 forms a complex with other proteins, such as FBXW8 and SKP1, to form an E3 ubiquitin ligase complex. This complex is responsible for the ubiquitination of target proteins, which are then recognized and degraded by the proteasome.
Clinical Significance[edit | edit source]
Mutations in the CUL7 gene have been associated with a rare genetic disorder known as 3-M syndrome. This syndrome is characterized by pre- and postnatal growth retardation, facial dysmorphism, and skeletal abnormalities. The disorder is inherited in an autosomal recessive manner, and individuals with mutations in CUL7 exhibit a range of clinical features associated with impaired growth and development.
Research has also suggested that CUL7 may play a role in cancer biology. Alterations in the expression or function of CUL7 have been observed in various types of cancer, indicating that it may have a role in tumorigenesis.
Interactions[edit | edit source]
CUL7 interacts with several other proteins to form functional complexes. Notably, it interacts with FBXW8, a member of the F-box protein family, which is involved in substrate recognition. CUL7 also interacts with SKP1, another component of the SCF (SKP1-CUL1-F-box protein) complex, which is a type of E3 ubiquitin ligase complex.
