Genome Walking is a method used in molecular biology to identify nucleotide sequences directly adjacent to a known sequence within the same DNA molecule. It is a technique used to isolate, sequence, and map unknown DNA regions connected to known sequences.
Overview[edit | edit source]
The process of Genome Walking involves the use of a known sequence to amplify the unknown regions of the DNA molecule. This is achieved through the use of polymerase chain reaction (PCR), a common method in molecular biology to make many copies of a specific DNA segment.
Methodology[edit | edit source]
The Genome Walking technique involves three main steps:
- Library Preparation: A genomic library is prepared by digesting the genomic DNA with a restriction enzyme. The fragments are then ligated to a known DNA sequence or 'adaptor' which will serve as a primer binding site in the PCR reaction.
- Primary PCR: The first round of PCR is performed using a gene-specific primer (GSP) that binds to the known sequence and a primer that binds to the adaptor sequence. This amplifies the region between the known sequence and the unknown sequence.
- Nested PCR: A second round of PCR is performed using a nested GSP that binds within the known sequence and a primer that binds to the adaptor. This amplifies a smaller region and increases the specificity of the reaction.

Genome Walking is used in various fields of genetics and biotechnology, including:
- Gene Mapping: It helps in mapping the genes and understanding the organization of the genes within the genome.
- Mutation Analysis: It is used to identify and study mutations in the genome.
- Cloning: It is used in cloning to identify the insertion sites of transgenes.
Limitations[edit | edit source]
While Genome Walking is a powerful tool, it has some limitations. It requires a known sequence for primer design, which may not always be available. It can also be time-consuming and may not always yield the desired results.
