Hooker County is a county in the U.S. state of Nebraska. As of the 2010 United States Census, the population was 736. Its county seat is Mullen. The county was formed in 1889 and named for Civil War General Joseph Hooker.

Hooker County is located in the Sandhills region of Nebraska, an area characterized by sand dunes and grass-covered prairies. The county has a total area of 721 square miles, of which 721 square miles is land and 0.4 square miles is water.

As of the 2010 United States Census, there were 736 people, 349 households, and 220 families residing in the county. The population density was 1.0 inhabitants per square mile. There were 505 housing units at an average density of 0.7 per square mile.
