TAB1
TAB1 or TGF-beta activated kinase 1/MAP3K7 binding protein 1 is a protein that in humans is encoded by the TAB1 gene. TAB1 is a critical component of the TGF-beta signaling pathway. It binds to TAK1 and aids in its activation upon the stimulation of TGF-beta.
Function[edit | edit source]
TAB1 is an activator of MAP3K7/TAK1, which is required for the IL-1 induced activation of NF-kappaB and JNK. This protein forms a kinase complex with TRAF6, MAP3K7 and TAB2; this complex is required for the activation of MAP3K7 following engagement of the Toll-like receptors. MAP3K7 activated by the binding of TAB1 to TRAF6, which is required for K63-linked polyubiquitination of TRAF6, and the subsequent activation of IKK and NF-kappaB.
Clinical significance[edit | edit source]
Mutations in TAB1 have been associated with congenital heart defects.
