ABI-007

ABI-007 is a formulation of the chemotherapy drug paclitaxel that is bound to albumin nanoparticles. It is commonly known by its trade name, Abraxane. ABI-007 is used in the treatment of various types of cancer, including breast cancer, non-small cell lung cancer, and pancreatic cancer.

Mechanism of Action[edit | edit source]

ABI-007 works by inhibiting the normal function of microtubules during cell division. Paclitaxel, the active ingredient, stabilizes microtubules and prevents their disassembly, which is necessary for cell division. This action leads to the inhibition of mitosis, resulting in cell death. The albumin-bound formulation allows for better solubility and delivery of the drug to the tumor site.

Clinical Uses[edit | edit source]

Breast Cancer[edit | edit source]

ABI-007 is approved for the treatment of metastatic breast cancer in patients who have failed combination chemotherapy or relapse within six months of adjuvant chemotherapy. It is often used when other treatments have not been effective.

Non-Small Cell Lung Cancer[edit | edit source]

In combination with carboplatin, ABI-007 is used as a first-line treatment for patients with locally advanced or metastatic non-small cell lung cancer who are not candidates for curative surgery or radiation therapy.

Pancreatic Cancer[edit | edit source]

ABI-007, in combination with gemcitabine, is approved for the treatment of metastatic pancreatic cancer. This combination has been shown to improve overall survival compared to gemcitabine alone.

Side Effects[edit | edit source]

Common side effects of ABI-007 include neutropenia, anemia, thrombocytopenia, peripheral neuropathy, nausea, vomiting, diarrhea, and alopecia. Serious side effects can include severe allergic reactions, pneumonitis, and sepsis.

Administration[edit | edit source]

ABI-007 is administered as an intravenous infusion. The dosage and schedule depend on the type of cancer being treated, the patient's overall health, and other factors.
