AM-4056

AM-4056 is a synthetic cannabinoid that is structurally related to other synthetic cannabinoids. It is a potent and selective agonist for the peripheral CB2 receptor, but has much lower affinity for the central CB1 receptor.

Chemistry[edit | edit source]

AM-4056 is a member of the AM series of cannabinoids, which are named after Alexandros Makriyannis, the scientist who first synthesized them. It is a derivative of CP 55,940, which is itself a derivative of cannabidiol, a naturally occurring cannabinoid found in cannabis.

Pharmacology[edit | edit source]

AM-4056 acts as a potent and selective agonist for the CB2 receptor, with a Ki value of 0.28 nM, making it one of the most potent CB2 agonists known. It has much lower affinity for the CB1 receptor, with a Ki value of 45.5 nM. This makes it over 160 times more selective for the CB2 receptor than the CB1 receptor.

Therapeutic potential[edit | edit source]

Due to its selectivity for the CB2 receptor, AM-4056 has potential therapeutic applications in conditions where activation of this receptor is beneficial, such as inflammatory diseases, pain, and cancer. However, as of 2021, it is still in the early stages of research and has not yet been tested in humans.
