An AMPA receptor positive allosteric modulator is a type of compound that enhances the activity of the AMPA receptor, a subtype of the ionotropic glutamate receptors in the central nervous system. These modulators bind to the receptor at a site distinct from the glutamate binding site, leading to an increase in the receptor's response to glutamate.

Mechanism of Action[edit | edit source]

AMPA receptor positive allosteric modulators work by binding to the allosteric site on the AMPA receptor. This binding induces a conformational change in the receptor, which increases its affinity for glutamate and/or prolongs the open state of the ion channel.
