Bone morphogenetic protein 1 (BMP1) is a protein that in humans is encoded by the BMP1 gene. Unlike other members of the bone morphogenetic protein family, BMP1 is not a growth factor but rather a metalloprotease enzyme that plays a crucial role in the formation of the extracellular matrix and the development of bone and cartilage.
Function[edit | edit source]
BMP1 is a member of the astacin family of zinc-dependent metalloproteases. It is involved in the processing of procollagen to collagen, which is a major component of the extracellular matrix. BMP1 cleaves the C-terminal propeptides of procollagen I, II, and III, facilitating their assembly into mature collagen fibrils. This process is essential for the structural integrity and function of connective tissues.
In addition to its role in collagen processing, BMP1 is also involved in the activation of other proteins, such as lysyl oxidase, which is important for the cross-linking of collagen and elastin fibers, further contributing to the strength and elasticity of connective tissues.
Clinical significance[edit | edit source]
Mutations in the BMP1 gene can lead to disorders of connective tissue, such as osteogenesis imperfecta, a condition characterized by brittle bones that are prone to fracture. Understanding the function of BMP1 and its role in collagen processing is important for developing therapeutic strategies for such conditions.

Research on BMP1 continues to explore its potential as a therapeutic target for diseases involving connective tissue defects. Studies are also investigating the broader role of BMP1 in tissue repair and regeneration, given its involvement in extracellular matrix formation.
