A chromosome puff or Balbiani ring is a region of a chromosome that has undergone chromatin decondensation and is actively engaged in transcription. These structures are typically observed in the polytene chromosomes of certain insects and are named after the Swedish scientist Edouard-Gerard Balbiani who first described them.

Chromosome puffs are visible as swollen regions along the length of a polytene chromosome. They occur in response to active gene transcription, where the chromatin fibers unwind to allow access to the underlying DNA. This process is facilitated by RNA polymerase and other transcription factors.
Formation and Function[edit | edit source]
The formation of a chromosome puff is a dynamic process that reflects the transcriptional activity of the cell. When a gene is activated for transcription, the chromatin in that region begins to unwind, forming a puff. The size and number of puffs can change depending on the cell's metabolic activity and developmental stage.
Chromosome puffs play a crucial role in gene expression. They allow for the amplification of specific genes, enabling the cell to rapidly produce large quantities of a particular protein or RNA molecule. This is especially important in cells that need to respond quickly to environmental changes or developmental cues.

Chromosome puffs have been extensively studied in the salivary glands of the fruit fly Drosophila melanogaster. These studies have provided valuable insights into the mechanisms of gene regulation and chromatin structure. Furthermore, the study of chromosome puffs has implications for understanding human diseases, such as cancer, where abnormal gene expression and chromatin remodeling are often observed.
