Ciclopirox Olamine is a synthetic antifungal agent used primarily in the treatment of dermatophytosis, onychomycosis, and other fungal infections of the skin and nails. It is available in various formulations, including creams, lotions, and nail lacquers.
Mechanism of Action[edit | edit source]
Ciclopirox Olamine exerts its antifungal effects by chelating trivalent cations such as Fe3+ and Al3+, which are essential for the enzymatic activity of fungal cells. This chelation disrupts the synthesis of ergosterol, a critical component of the fungal cell membrane, leading to increased cell permeability and ultimately cell death. Additionally, Ciclopirox has been shown to inhibit mitochondrial electron transport and DNA repair processes in fungal cells.
Pharmacokinetics[edit | edit source]
Ciclopirox Olamine is applied topically and has negligible systemic absorption. It binds extensively to plasma proteins, with a binding rate of 94-97%. The drug is metabolized in the liver and has an elimination half-life of approximately 1.7 hours. It is primarily excreted in the urine.
