FZD3 (Frizzled Class Receptor 3) is a protein that in humans is encoded by the FZD3 gene. This gene is a member of the frizzled gene family, which is involved in the Wnt signaling pathway. The Wnt signaling pathway plays a critical role in embryonic development, cell proliferation, migration, and differentiation. The FZD3 receptor is one of several receptors that can bind Wnt proteins and initiate the signaling cascade.

Function[edit | edit source]

FZD3 is a G protein-coupled receptor that is activated by binding to Wnt proteins. Upon activation, FZD3 initiates a cascade of cellular signals that lead to various biological outcomes depending on the cellular context and the specific Wnt ligand involved. In the canonical Wnt signaling pathway, FZD3 activation leads to the stabilization and accumulation of beta-catenin in the cytoplasm and its subsequent translocation into the nucleus where it influences the expression of target genes.

In addition to its role in the canonical pathway, FZD3 is also involved in non-canonical Wnt signaling pathways, including the planar cell polarity (PCP) pathway, which is important for the regulation of cell shape, movement, and organization in tissues.

Alterations in the FZD3 gene or its signaling pathway have been implicated in various human diseases and conditions. Abnormal FZD3 signaling has been associated with cancer, where it can influence tumor growth, metastasis, and resistance to therapy. In neuroscience, FZD3 signaling is important for the development of the nervous system, and mutations in the FZD3 gene have been linked to neurodevelopmental disorders and psychiatric conditions such as schizophrenia.

The FZD3 gene is located on human chromosome 8. Variants and mutations in this gene can affect the normal signaling function of the FZD3 receptor, leading to altered cellular responses to Wnt proteins.
