Noradrenergic and specific serotonergic antidepressants (NaSSAs) are a class of antidepressant drugs used primarily in the treatment of depression. They are also used in the treatment of anxiety disorders, obsessive-compulsive disorder (OCD), and occasionally for other mental health conditions. NaSSAs work by modulating the neurotransmitters norepinephrine and serotonin in the brain, which are believed to be involved in the regulation of mood and behavior.

Mechanism of Action[edit | edit source]

NaSSAs act by antagonizing alpha-2 adrenergic receptors, which increases the release of norepinephrine and serotonin in the brain. Unlike many other antidepressants, NaSSAs selectively target specific serotonin receptors, such as the 5-HT2A and 5-HT2C receptors, while having minimal effects on the reuptake of serotonin. This selective mechanism is thought to contribute to the antidepressant and anxiolytic effects of NaSSAs with fewer side effects related to serotonin, such as sexual dysfunction and gastrointestinal disturbances.

NaSSAs are primarily used in the treatment of major depressive disorder (MDD). Due to their unique mechanism of action, NaSSAs can also be effective in treating anxiety disorders and are sometimes used as an adjunct therapy for schizophrenia and other psychiatric conditions.

The side effects of NaSSAs can vary but may include drowsiness, weight gain, dry mouth, constipation, and blurred vision.
