Pou domain

Pou domain is a protein domain that is found in a family of transcription factors. These transcription factors play a crucial role in the development and function of various organisms, including humans. The name "Pou" is derived from the first letters of "Pituitary specific Pit-1", "Octamer transcription factor-1" and "Unc-86", the three proteins in which the domain was first identified.

Structure[edit | edit source]

The Pou domain is characterized by a highly conserved region of approximately 150 amino acids. This region is responsible for DNA binding and is composed of two subdomains: the POU-specific (POUs) domain and the POU homeodomain (POUh). The POUs domain consists of about 75 amino acids, while the POUh domain is slightly larger, with around 60 amino acids. The two subdomains are connected by a flexible linker that allows for considerable spatial freedom, enabling the POU domain to bind to various DNA sequences.

Function[edit | edit source]

Transcription factors containing the POU domain are involved in a wide range of biological processes. They regulate the expression of genes by binding to specific DNA sequences, thereby controlling the transcription of these genes into RNA. This regulation plays a crucial role in cell differentiation, development, and metabolic processes. In humans, POU domain proteins have been implicated in various diseases, including cancer and neurodegenerative disorders.

Classification[edit | edit source]

Based on sequence similarities, POU domain proteins can be classified into six classes, from POU1 to POU6. Each class is further divided into subclasses, which are designated by a letter. For example, the POU2 class includes the subclasses POU2F1 and POU2F2.

  • Pit-1: A POU1 class protein that is essential for the development of the pituitary gland in mammals.
  • Oct-1 and Oct-2: POU2 class proteins that are involved in the immune response in mammals.
  • Unc-86: A POU4 class protein that plays a key role in the development of the nervous system in the nematode Caenorhabditis elegans.
