Reporter gene

From WikiMD's Food, Medicine & Wellness Encyclopedia

Reporter gene is a gene that researchers use in molecular biology to study the activity of a particular gene promoter. The reporter gene is attached to the gene of interest's regulatory sequence. When the gene of interest is transcribed, the reporter gene is also transcribed. The reporter gene's product is easy to detect and measure, so it can be used to determine when and where the gene of interest is being expressed.

Function[edit | edit source]

The reporter gene is used to indicate if a certain gene has been taken up by or expressed in the cell or organism population. The reporter gene can be placed next to the gene of interest in a plasmid used for transformation. The reporter gene's expression indicates that the gene of interest is being expressed. The reporter gene can also be attached to the gene of interest in a gene fusion to study the gene's expression and localization.

Types of reporter genes[edit | edit source]

There are several types of reporter genes that can be used, depending on the specific needs of the study. Some of the most common reporter genes include:

  • Beta-galactosidase: This enzyme is often used in bacteria. It can cleave lactose into glucose and galactose, and its activity can be measured by a colorimetric assay.
  • Green fluorescent protein (GFP): This protein glows green when exposed to blue light. It is often used in eukaryotic cells.
  • Luciferase: This enzyme is responsible for the glow of fireflies. It can be used in many types of organisms and can be measured using a luminometer.
  • Chloramphenicol acetyltransferase (CAT): This enzyme can be used in many types of cells. It acetylates chloramphenicol, and its activity can be measured by an immunoassay.

Applications[edit | edit source]

Reporter genes are used in a variety of applications in molecular biology. They can be used to study gene expression, to track the spread of a virus in a population, to determine the effectiveness of a drug, or to study the function of a particular gene.
