Transmembrane Protease, Serine 6 (TMPRSS6) is a serine protease enzyme that in humans is encoded by the TMPRSS6 gene. TMPRSS6 plays a crucial role in iron homeostasis and erythropoiesis, the process of producing red blood cells. It is a type II transmembrane serine protease that is primarily expressed in the liver. TMPRSS6 is involved in the regulation of hepcidin, a key hormone in the control of iron absorption and distribution in the body.

Function[edit | edit source]

TMPRSS6 cleaves and inactivates hepcidin, a peptide hormone produced by the liver that plays a central role in regulating iron metabolism. By inactivating hepcidin, TMPRSS6 promotes increased iron absorption from the diet and release from body stores. This action is critical in maintaining adequate iron levels for erythropoiesis and preventing conditions such as iron deficiency anemia and anemia of chronic disease, where iron availability is limited.

Genetic Variations and Disease Association[edit | edit source]

Mutations in the TMPRSS6 gene are associated with iron-refractory iron deficiency anemia (IRIDA), a rare hereditary disease characterized by iron deficiency anemia that does not respond to oral iron supplementation and only partially responds to parenteral iron therapy. Patients with IRIDA have inappropriately high levels of hepcidin for their iron status, which is believed to be due to the lack of TMPRSS6 activity on hepcidin regulation.

Clinical Significance[edit | edit source]

Understanding the role of TMPRSS6 in iron metabolism has significant clinical implications. It offers potential targets for therapeutic intervention in conditions of iron overload, such as hemochromatosis, and in conditions of iron deficiency, such as certain types of anemia. Pharmacological modulation of TMPRSS6 activity could provide a novel approach to managing these conditions.

Current research is focused on further elucidating the mechanism of action of TMPRSS6 and its interaction with other proteins involved in iron metabolism. Additionally, studies are exploring the potential of TMPRSS6 as a therapeutic target for the treatment of iron-related disorders.
