Aquaporin 6 (AQP6) is a protein that in humans is encoded by the AQP6 gene. It belongs to the aquaporin family, a group of small, integral membrane proteins that facilitate water transport across biological membranes. AQP6 has been identified primarily in the kidney, specifically within the intracellular vesicles of the acid-secreting intercalated cells of the renal collecting duct. Its expression and localization suggest a role in water and possibly anion transport in the kidney, contributing to the regulation of acid-base balance and fluid homeostasis.

AQP6 is an atypical member of the aquaporin family. Unlike other aquaporins that primarily facilitate passive water transport, AQP6 can function as an anion channel under certain conditions. It has been observed to conduct anions such as nitrate when activated by low pH or mercurial agents, indicating a potential role in cellular processes that require rapid changes in ion composition. However, the physiological significance of this anion transport capability is still under investigation.

In the kidney, AQP6 is thought to participate in the fine-tuning of urine concentration and acidification. Its presence in acid-secreting cells suggests a role in the reabsorption or secretion of water and ions, contributing to the maintenance of acid-base balance and osmotic gradients.

Gene and Expression[edit | edit source]

The AQP6 gene is located on chromosome 12 in humans. Its expression is relatively restricted compared to other aquaporins, with the highest levels found in the kidney. Within the kidney, AQP6 is localized to the intracellular vesicles of intercalated cells in the collecting duct, a key site for the regulation of urine concentration and pH.

Clinical Significance[edit | edit source]

While the specific physiological functions of AQP6 are still being elucidated, alterations in its expression or function could potentially impact renal water handling and ion balance. However, as of now, there are no direct associations between AQP6 and specific kidney diseases. Further research may reveal whether mutations in the AQP6 gene or dysregulation of AQP6 protein expression contribute to renal pathologies or other diseases.

Ongoing research aims to clarify the precise physiological role of AQP6 in the kidney and other tissues. Studies are also exploring the regulation of AQP6 expression and trafficking within cells, as well as its potential interactions with other proteins.
