Carbonic Anhydrase 7 (CA7) is an enzyme that belongs to the carbonic anhydrase family. It plays a crucial role in catalyzing the reversible hydration of carbon dioxide (CO2) to bicarbonate (HCO3-) and a proton (H+). This enzyme is encoded by the CA7 gene, which is located on chromosome 16 in humans.
Structure[edit]
The structure of Carbonic Anhydrase 7 consists of a single polypeptide chain, which folds into a compact globular shape. It contains a zinc ion at its active site, which is essential for its catalytic activity. The enzyme has a molecular weight of approximately 30 kDa.
Function[edit]
Carbonic Anhydrase 7 is primarily expressed in the brain, specifically in the cerebellum and hippocampus. It is involved in maintaining the acid-base balance in these regions, as well as regulating the production of cerebrospinal fluid. Additionally, CA7 has been found to play a role in neuronal development and synaptic plasticity.
The catalytic activity of Carbonic Anhydrase 7 is crucial for the transport of carbon dioxide and bicarbonate across cell membranes. By converting CO2 into HCO3- and H+, it helps maintain the pH balance within cells and tissues. This process is particularly important in tissues with high metabolic activity, such as the brain.
Clinical Significance[edit]
Mutations in the CA7 gene have been associated with certain neurological disorders, including epilepsy and ataxia. These mutations can disrupt the normal function of Carbonic Anhydrase 7, leading to imbalances in pH regulation and impaired neuronal development.
Furthermore, Carbonic Anhydrase 7 has been identified as a potential therapeutic target for the treatment of glaucoma. Inhibition of this enzyme has shown promise in reducing intraocular pressure, which is a major risk factor for glaucoma.
