Embedding is a process in histology where tissue samples are encapsulated in a solid medium, such as paraffin wax or plastic resin, to allow thin sections to be cut and mounted on a microscope slide for examination. This process is crucial in the preparation of tissue samples for microscopy.
Process[edit | edit source]
The embedding process begins with the fixation of the tissue sample to preserve its structure. This is typically done using a fixative such as formalin. The tissue is then dehydrated, usually through a series of alcohol baths, to remove water. The dehydrated tissue is then cleared, which involves replacing the alcohol with a substance that is miscible with both alcohol and the embedding medium. The cleared tissue is then infiltrated with the embedding medium, which is subsequently hardened to form a block. The tissue can then be sectioned using a microtome.
Embedding Mediums[edit | edit source]
There are several different types of embedding mediums that can be used, each with their own advantages and disadvantages.
- Paraffin wax: This is the most commonly used embedding medium. It is easy to use and provides good preservation of tissue structure. However, it does not provide as fine a level of detail as some other mediums.
- Plastic resins: These provide a higher level of detail than paraffin wax, but are more difficult to use. They are typically used for electron microscopy.
- Frozen sections: In some cases, tissue may be frozen and sectioned without the use of an embedding medium. This is typically done for rapid diagnosis during surgery.

It is a crucial step in the preparation of tissue samples for histological examination, which is used in the diagnosis of many diseases, including cancer.
