Gladys Marie Deacon was born in Paris, France in 1881. She was the daughter of Edward Deacon, a wealthy American businessman, and his wife Florence. Gladys grew up in a privileged environment and received a good education.

In 1899, Gladys married the 9th Duke of Marlborough, George Spencer-Churchill. Following her marriage, she became known as the Duchess of Marlborough. The couple had a tumultuous relationship, and they eventually separated.
