Interleukin-6 receptor (IL-6R) is a type of protein and a part of the immune system that plays a critical role in mediating the effects of interleukin-6 (IL-6), a cytokine involved in various physiological and pathological processes. The IL-6 receptor is essential for the immune response, inflammation, and the regulation of metabolic, regenerative, and neural processes.

Structure[edit | edit source]

The IL-6 receptor is composed of two main subunits: an alpha chain (IL-6Rα) and a beta chain (gp130). The alpha chain is responsible for binding IL-6 with high affinity, while the beta chain, gp130, is shared with other cytokine receptors and is crucial for signal transduction. The IL-6R exists in two forms: a membrane-bound form and a soluble form (sIL-6R). The soluble form can bind to IL-6 and interact with gp130 on the cell surface, thus enabling IL-6 signal transduction in cells that do not express the membrane-bound receptor.

Function[edit | edit source]

The primary function of the IL-6 receptor is to mediate the effects of IL-6, which include the stimulation of immune responses, inflammation, hematopoiesis (formation of blood cells), and the regulation of metabolic processes. IL-6 signaling through its receptor can lead to the activation of various signaling pathways, including the JAK/STAT pathway, which plays a key role in the regulation of gene expression.

Clinical Significance[edit | edit source]

The IL-6/IL-6R signaling pathway is implicated in the pathogenesis of several diseases, including rheumatoid arthritis, multiple myeloma, and Castleman's disease. Abnormal activation of IL-6 signaling can lead to chronic inflammation and autoimmunity. As a result, targeting the IL-6 receptor has emerged as a therapeutic strategy in treating these conditions. Tocilizumab, a monoclonal antibody against IL-6R, has been approved for the treatment of rheumatoid arthritis and other inflammatory diseases.

Research on the IL-6 receptor continues to uncover its role in health and disease. Studies are exploring its involvement in metabolic syndrome, cardiovascular diseases, and cancer.
