Jefferson County is a county located in the U.S. state of Pennsylvania. As of the 2020 census, the population was 42,076. The county seat is Brookville. The county was created on October 1, 1830, and was named in honor of Thomas Jefferson, the author of the Declaration of Independence and the third President of the United States.

According to the United States Census Bureau, the county has a total area of 657 square miles (1,700 km²), of which 652 square miles (1,690 km²) is land and 5.0 square miles (13 km²) (0.8%) is water.
