UFSP2 (Ubiquitin-Fold Modifier 1-Specific Protease 2) is a protein encoded by the UFSP2 gene in humans. This protein is involved in the processing of UFM1, a ubiquitin-like protein, and plays a crucial role in the ubiquitin-proteasome system.

UFSP2 is a cysteine protease that specifically cleaves the C-terminal extension of the ubiquitin-fold modifier 1 (UFM1) precursor, thereby generating the mature form of UFM1. This processing is essential for the conjugation of UFM1 to its target proteins, a process known as UFMylation. UFMylation is implicated in various cellular processes, including protein quality control, endoplasmic reticulum stress response, and cell cycle regulation.

The UFSP2 protein contains a conserved cysteine protease domain, which is responsible for its enzymatic activity. The structure of UFSP2 allows it to recognize and bind to the UFM1 precursor, facilitating its cleavage and maturation.

Mutations in the UFSP2 gene have been associated with various human diseases. For instance, certain mutations can lead to neurodegenerative disorders and skeletal dysplasia.
