ADAM10 (A Disintegrin And Metalloproteinase 10) is a protein that in humans is encoded by the ADAM10 gene. It is a member of the ADAM (A Disintegrin And Metalloproteinase) protein family. ADAM10 plays a crucial role in the Notch signaling pathway, which is involved in cell-cell communication and regulates a broad spectrum of cell-fate decisions.

Function[edit | edit source]

ADAM10 is a type I transmembrane protein and is a functional enzyme that has both protease and integrin binding activities. It is involved in the processing of numerous proteins, including growth factors, cytokines, and cell adhesion molecules. ADAM10 is also responsible for the cleavage of amyloid precursor protein (APP), which results in the production of amyloid beta, a protein fragment that accumulates in the brains of individuals with Alzheimer's disease.

Clinical significance[edit | edit source]

Alterations in the expression or activity of ADAM10 have been implicated in a variety of diseases, including cancer, inflammatory diseases, and neurodegenerative diseases. In cancer, ADAM10 is often overexpressed and contributes to tumor progression by enhancing cell proliferation, invasion, and angiogenesis. In inflammatory diseases, ADAM10 can promote inflammation by shedding pro-inflammatory cytokines and growth factors. In neurodegenerative diseases, such as Alzheimer's disease, increased activity of ADAM10 can lead to the accumulation of amyloid beta, contributing to the pathogenesis of the disease.

Research on ADAM10 has focused on understanding its role in health and disease, and on developing therapeutic strategies to modulate its activity. Several inhibitors of ADAM10 have been developed and are being tested in preclinical and clinical trials for the treatment of various diseases.
