Velpeau was born in Brèches, a small village in the Indre-et-Loire department of France. He initially trained as a pharmacist before pursuing a career in medicine. Velpeau studied at the University of Tours and later at the University of Paris, where he was mentored by prominent surgeons of the time.

Velpeau became a professor of clinical surgery at the University of Paris and was appointed to the prestigious position of chair of clinical surgery at the Hôpital de la Charité. His career was marked by numerous advancements in surgical practices and education.
