CRLF2 (Cytokine Receptor-Like Factor 2) is a protein that in humans is encoded by the CRLF2 gene. This protein is a type I cytokine receptor and is part of the hematopoietic receptor family. It plays a crucial role in the development and function of the immune system.

Structure[edit | edit source]

The CRLF2 protein is composed of several domains that are characteristic of cytokine receptors. It has an extracellular domain that binds to its ligand, a single transmembrane domain, and an intracellular domain that is involved in signal transduction. The gene encoding CRLF2 is located on the pseudoautosomal region of the X chromosome and Y chromosome.

Function[edit | edit source]

CRLF2 forms a receptor complex with the Interleukin-7 receptor alpha (IL-7Rα) chain. This complex is involved in the signaling pathways that regulate the development and function of B cells and T cells. CRLF2 is also known to interact with the Thymic stromal lymphopoietin (TSLP), which is important for the maturation of dendritic cells and the activation of T helper cells.

Clinical Significance[edit | edit source]

Mutations and overexpression of the CRLF2 gene have been implicated in various types of leukemia, particularly acute lymphoblastic leukemia (ALL). These genetic alterations can lead to abnormal signaling pathways that promote the proliferation and survival of leukemic cells. CRLF2 abnormalities are often associated with poor prognosis in ALL patients.

Ongoing research is focused on understanding the precise mechanisms by which CRLF2 contributes to leukemia and other immune-related disorders. Studies are also exploring potential therapeutic targets within the CRLF2 signaling pathway to develop new treatments for these conditions.
