CYP11B2
CYP11B2 is an enzyme that plays a crucial role in the biosynthesis of aldosterone and other steroid hormones. It is encoded by the CYP11B2 gene in humans. This enzyme is part of the cytochrome P450 family, which is involved in the synthesis and metabolism of various molecules and chemicals within cells. CYP11B2 specifically catalyzes the final steps of aldosterone production in the adrenal cortex, a process essential for regulating blood pressure and electrolyte balance.

CYP11B2 is primarily found in the zona glomerulosa of the adrenal cortex. It converts 11-deoxycorticosterone (DOC) to corticosterone and then to aldosterone through a series of oxidation and hydroxylation steps. This process is critical for the maintenance of salt and water homeostasis in the body, influencing blood pressure and potassium levels. Aldosterone acts on the kidneys to increase sodium reabsorption and potassium excretion, which in turn regulates blood pressure.

The expression of the CYP11B2 gene is regulated by various factors, including angiotensin II, potassium levels, and the adrenocorticotropic hormone (ACTH). Angiotensin II, in particular, is a key regulator in response to low blood pressure or low sodium levels in the blood, stimulating aldosterone synthesis to restore balance.
Clinical Significance[edit | edit source]
Mutations or dysregulation in the CYP11B2 gene can lead to disorders related to aldosterone production. One such condition is primary aldosteronism, also known as Conn's syndrome, where excessive aldosterone production leads to hypertension and hypokalemia. Conversely, inadequate function of CYP11B2 can result in hypoaldosteronism, characterized by low blood pressure and hyperkalemia.
In addition to its role in disease, CYP11B2 is a target for certain antihypertensive drugs. Inhibitors of this enzyme can reduce aldosterone levels, offering a therapeutic approach for conditions like hypertension and heart failure.
