John Strother Griffin (1816–1898) was an American physician and a prominent figure in the early history of Los Angeles, California. He played a significant role in the development of the city's medical infrastructure and was instrumental in the establishment of several key institutions.

Griffin was born on June 25, 1816, in Falmouth, Virginia. He pursued his medical education at the University of Pennsylvania, where he graduated with a degree in medicine in 1837. Following his graduation, Griffin served as an assistant surgeon in the United States Army.

During the Mexican-American War, Griffin was stationed in California and served as a surgeon with the Army of the West. His service in the war brought him to the region that would later become his home.
