Kallikrein-related peptidase 9 (KLK9) is a member of the kallikrein subfamily of serine proteases, which are enzymes that cleave peptide bonds in proteins. KLK9 is encoded by the KLK9 gene located on chromosome 19q13.3, a region known for containing a cluster of kallikrein genes.
Structure[edit | edit source]
The KLK9 gene is composed of five exons and encodes a preproprotein that is processed to generate the mature enzyme. The mature KLK9 protein consists of approximately 250 amino acids and contains the characteristic serine protease catalytic triad: histidine, aspartate, and serine.
Function[edit | edit source]
KLK9 is involved in various physiological processes, including skin desquamation, semen liquefaction, and possibly in the regulation of blood pressure. Like other kallikreins, KLK9 is thought to play a role in the degradation of extracellular matrix components, which is important in tissue remodeling and repair.
Expression[edit | edit source]
KLK9 is expressed in a variety of tissues, with the highest levels found in the central nervous system, ovary, and testis. Its expression is regulated by steroid hormones, particularly androgens and estrogens, suggesting a role in reproductive physiology.
Clinical Significance[edit | edit source]
Altered expression of KLK9 has been observed in several types of cancer, including breast cancer and ovarian cancer. It is being studied as a potential biomarker for cancer diagnosis and prognosis. Elevated levels of KLK9 in serum or tissue samples may correlate with disease progression or response to therapy.

Ongoing research is focused on understanding the precise biological functions of KLK9 and its potential as a therapeutic target. Studies are also investigating the role of KLK9 in neurodegenerative diseases and its involvement in inflammatory processes.
