Oni Blackstock was born and raised in the United States. She pursued her undergraduate studies at Harvard University, where she earned a Bachelor of Arts degree. She then attended Harvard Medical School, where she obtained her Doctor of Medicine (MD) degree. Following medical school, Blackstock completed her residency in internal medicine at Montefiore Medical Center in New York City.

After completing her residency, Blackstock joined the faculty at Albert Einstein College of Medicine and Montefiore Medical Center.
