Pimobendan


Pimobendan is a type of phosphodiesterase inhibitor and calcium sensitizer that is primarily used in the treatment of heart failure in dogs. It is a non-sympathomimetic, non-glycoside inotropic drug that also has vasodilatory properties.

Pharmacology[edit | edit source]

Pimobendan works by inhibiting the function of phosphodiesterase III and increasing the sensitivity of cardiac muscle to calcium. This dual action results in increased contractility of the heart (positive inotropy) and vasodilation. The inhibition of phosphodiesterase III causes an increase in the levels of cyclic AMP, which in turn enhances calcium influx into the cardiac muscle cells, leading to increased contractility. The calcium sensitizing effect is achieved by a different mechanism, which involves binding to troponin C in the cardiac muscle cells and increasing their sensitivity to calcium.

Clinical Use[edit | edit source]

Pimobendan is used in the management of congestive heart failure in dogs, particularly those caused by dilated cardiomyopathy and mitral valve disease. It has been shown to improve the quality of life and extend survival time in dogs with these conditions. Pimobendan is typically used in combination with other medications such as diuretics, ACE inhibitors, and beta blockers.

Side Effects[edit | edit source]

The most common side effects of pimobendan include loss of appetite, lethargy, diarrhea, and difficulty breathing. In rare cases, it can cause an abnormal heart rhythm.

Dosage and Administration[edit | edit source]

Pimobendan is available in tablet form and is typically administered orally twice a day. The dosage is based on the weight of the dog and the severity of the heart condition.
