Umespirone

Umespirone is a serotonin 1A receptor agonist that was under development for the treatment of depression and anxiety but was never marketed. It is structurally related to the atypical antipsychotic buspirone and the antidepressant ipsapirone.

Pharmacology[edit | edit source]

Umespirone acts as a partial agonist of the 5-HT1A receptor. This action is thought to mediate its anxiolytic and antidepressant effects. Umespirone also has weak affinity for the dopamine D2 receptor and the alpha-1 adrenergic receptor.

Development[edit | edit source]

Umespirone was under development by Upjohn (now part of Pfizer) in the 1980s and 1990s. It reached phase II clinical trials for depression and anxiety but was discontinued and never marketed.
