Berotralstat hydrochloride

Berotralstat Hydrochloride is a medication used in the treatment of hereditary angioedema, a rare genetic disorder characterized by episodes of severe swelling. It is marketed under the brand name Orladeyo by BioCryst Pharmaceuticals.

Mechanism of Action[edit | edit source]

Berotralstat hydrochloride works by inhibiting plasma kallikrein, an enzyme that plays a key role in the inflammatory response that leads to swelling in hereditary angioedema. By blocking this enzyme, berotralstat hydrochloride can prevent or reduce the severity of swelling episodes.

Uses[edit | edit source]

Berotralstat hydrochloride is approved for use in adults and children 12 years of age and older with hereditary angioedema. It is taken orally once daily.

Side Effects[edit | edit source]

Common side effects of berotralstat hydrochloride include abdominal pain, vomiting, diarrhea, back pain, and heartburn. Serious side effects can include liver damage.

History[edit | edit source]

Berotralstat hydrochloride was approved by the Food and Drug Administration (FDA) in December 2020, making it the first oral medication approved for the prevention of attacks in hereditary angioedema patients.
