Brooke County is a county located in the U.S. state of West Virginia. As of the 2020 census, the population was 21,106. Its county seat is Wellsburg. The county was formed in 1797 from parts of Ohio County and named in honor of Robert Brooke, Governor of Virginia from 1794 to 1796.

According to the United States Census Bureau, the county has a total area of 93 square miles (240 km²), of which 89 square miles (230 km²) is land and 4.3 square miles (11 km²) (4.3%) is water. The county is part of the Weirton–Steubenville metropolitan area.

As of the 2020 census, there were 21,106 people, 9,000 households, and 5,800 families residing in the county. The population density was 237 people per square mile (91/km²). There were 10,000 housing units at an average density of 112 per square mile (43/km²).

Brooke County is served by the Brooke County Schools system. The county is home to Bethany College, a private liberal arts college in Bethany.
