MST1R, also known as the Macrophage Stimulating 1 Receptor or RON tyrosine kinase, is a protein that in humans is encoded by the MST1R gene. This receptor is a member of the receptor tyrosine kinase (RTK) family, which plays a critical role in the regulation of cellular processes including metabolism, growth, and differentiation. MST1R is particularly significant in the context of immunology and cancer biology, where it influences macrophage activation and is implicated in the progression of various cancers.

Function[edit | edit source]

MST1R is activated by its ligand, Macrophage Stimulating Protein (MSP), leading to a cascade of downstream signaling events that promote cellular proliferation, survival, and motility. This receptor is predominantly expressed in cells of epithelial origin and is involved in the regulation of inflammatory responses, tissue repair, and homeostasis. In the immune system, MST1R's activation enhances the phagocytic ability of macrophages and contributes to the resolution of inflammation.

Clinical Significance[edit | edit source]

The aberrant expression or activation of MST1R has been linked to the development and progression of several types of cancers, including breast cancer, colorectal cancer, and lung cancer. Its role in promoting tumor growth, metastasis, and drug resistance makes MST1R a potential target for cancer therapy. Inhibitors of MST1R are currently being explored as therapeutic agents in preclinical and clinical trials, aiming to block its signaling pathway and thereby inhibit tumor progression.

The MST1R gene is located on chromosome 3 (3p21.3) in humans. Mutations in this gene, leading to the overexpression or constitutive activation of the MST1R protein, have been identified in various cancers. These genetic alterations contribute to the oncogenic properties of MST1R, including enhanced cell survival, proliferation, and migration.

Research on MST1R continues to uncover its complex role in both normal physiological processes and disease states.
