Sodium-hydrogen exchange regulatory cofactor 2 (also known as NHERF2, SLC9A3R2 or EBP50L1) is a protein that in humans is encoded by the SLC9A3R2 gene. This protein is a member of the NHERF family, and plays a crucial role in the regulation of ion transport and signal transduction pathways.

Function[edit | edit source]

NHERF2 is a PDZ domain-containing protein. PDZ domains are protein-protein interaction modules that bind to the C-termini of target proteins. NHERF2 is known to bind to a variety of proteins, including GPCRs, ion channels, and other signaling proteins. This binding helps to regulate the function and localization of these proteins.

NHERF2 is also involved in the regulation of sodium-hydrogen exchangers, which are important for maintaining pH homeostasis in cells. By binding to these exchangers, NHERF2 can influence their activity and thus affect the pH balance within cells.

Clinical significance[edit | edit source]

Mutations in the SLC9A3R2 gene have been associated with various diseases, including hypertension, kidney disease, and certain types of cancer. Further research is needed to fully understand the role of NHERF2 in these conditions.
