Fibronectin type III domain-containing protein 5 (FNDC5) is a protein that in humans is encoded by the FNDC5 gene. FNDC5 is a membrane protein that is involved in various biological processes, including the regulation of energy metabolism, inflammation, and insulin sensitivity. It has gained significant attention due to its role in the secretion of irisin, a hormone that is believed to drive the conversion of white fat cells into brown fat cells, thus playing a crucial role in the process of thermogenesis and obesity management.

FNDC5 is composed of a signal peptide, a fibronectin type III domain, and a hydrophobic region that anchors it to the membrane. The fibronectin type III domain is a common structural motif found in various proteins and is involved in protein-protein interactions.

The primary function of FNDC5 is its role in the secretion of irisin. Irisin is produced when the FNDC5 precursor is cleaved, and it is released into the circulation. Irisin has been shown to have several beneficial effects on metabolic diseases and energy expenditure, making FNDC5 a potential target for the treatment of diabetes, obesity, and other metabolic disorders.

In addition to its metabolic effects, FNDC5/irisin has been implicated in the regulation of neuronal development and cognition, suggesting a broader physiological role beyond metabolism.
