LB buffer (Lysogeny Broth buffer) is a nutrient-rich medium primarily used for the growth of bacteria, particularly Escherichia coli (E. coli). It is a crucial component in molecular biology, microbiology, and genetic engineering for the cultivation of bacterial cultures.

LB buffer typically consists of the following components:
- Tryptone: A source of amino acids and peptides, derived from the enzymatic digestion of casein.
- Yeast Extract: Provides vitamins, particularly B-complex vitamins, and other growth factors.
- Sodium Chloride (NaCl): Maintains osmotic balance and provides essential ions for cellular processes.
The standard formulation of LB buffer is:

- LB Broth (Miller): Contains 10 g/L NaCl.
- LB Broth (Lennox): Contains 5 g/L NaCl, used for more sensitive applications.
- LB Broth (Luria): Contains 0.5 g/L NaCl, used for specific experimental conditions.

LB buffer is widely used in:
- Bacterial Culture: It supports the rapid growth of E. coli and other bacteria, making it ideal for cloning and expression studies.
- Plasmid DNA Preparation: LB buffer is used to grow bacterial cultures for plasmid DNA extraction.
- Protein Expression: It is used in recombinant protein production, where bacteria are transformed with plasmids carrying the gene of interest.
Preparation[edit | edit source]
To prepare LB buffer:
1. Dissolve the required amounts of tryptone, yeast extract, and sodium chloride in distilled water. 2. Adjust the pH to 7.0 using NaOH or HCl if necessary. 3. Autoclave the solution at 121°C for 15 minutes to sterilize. 4. Cool the solution to room temperature before use.
Storage[edit | edit source]
LB buffer can be stored at room temperature or refrigerated. It should be kept in a sterile container to prevent contamination.
