Zarnestra

Zarnestra (also known as Tipifarnib) is a farnesyltransferase inhibitor that is being studied in the treatment of cancer. It is a type of antineoplastic agent.

Mechanism of Action[edit | edit source]

Zarnestra works by inhibiting the enzyme farnesyltransferase, an important player in the Ras signaling pathway. This pathway is often overactive in cancer cells, leading to uncontrolled cell growth and division. By inhibiting farnesyltransferase, Zarnestra prevents the activation of the Ras protein, thereby slowing or stopping the growth of cancer cells.

Clinical Trials[edit | edit source]

Zarnestra has been studied in a number of clinical trials for various types of cancer. These include acute myeloid leukemia (AML), non-Hodgkin lymphoma (NHL), and colorectal cancer. While results have been mixed, some studies have shown promise, particularly in patients with certain genetic mutations.

Side Effects[edit | edit source]

Like all chemotherapy drugs, Zarnestra can cause side effects. These may include nausea, vomiting, diarrhea, and fatigue. More serious side effects can include neutropenia (a decrease in the number of white blood cells), thrombocytopenia (a decrease in the number of platelets), and anemia (a decrease in the number of red blood cells).
