Vatanidipine is a calcium channel blocker of the dihydropyridine class, primarily used in the management of hypertension and angina pectoris. It functions by inhibiting the influx of calcium ions through L-type calcium channels, leading to vasodilation and a subsequent decrease in blood pressure.

Vatanidipine selectively inhibits L-type calcium channels in vascular smooth muscle and cardiac muscle. This inhibition prevents calcium ions from entering cells, which is crucial for muscle contraction. By reducing intracellular calcium, vatanidipine causes relaxation of vascular smooth muscle, leading to vasodilation and reduced peripheral resistance.

Vatanidipine is well absorbed from the gastrointestinal tract when administered orally. It has a high bioavailability and is extensively bound to plasma proteins (approximately 95%). The drug undergoes hepatic metabolism, primarily via the cytochrome P450 enzyme system, and is excreted mainly in the urine. The elimination half-life of vatanidipine ranges from 5 to 8 hours, allowing for once or twice daily dosing.
